What game is it first (and is it a special edition made to play on Vista?)
When you try to load are there any error messages?
When it doesnt load what happens, does it go to a black screen like its going to go into it and then flits back out to Windows?
Solutions -
Right click desktop shortcut, go to Properties then Compatibility and change to XP or 2000 or 98 (go in that order)
If that doesnt work attempt a complete uninstallation (uninstall using Add/Remove, then delete any program folders then reset) and install it again
Before posting back (unless you have real error messages) try the stuff above

Right click on the game shortcut .exe and select 'run as adminstrator'. Make any difference?
