I believe, the 4-1 will keep you're power in mid-range, and 4-2-1 will move it higher in you're range.
It depends how you're engine is set-up, which you want.

I've always wondered this myself. I thought maybe they were the same thing but people just word them differently.

Physically, a 4-2-1 goes from 4 headers (the 'pipes' that come from the flange), into 2 then the two go into one, which then goes onto the exhaust. If that's described well enough.
A 4-1 just misses out the middle stage and the 4 'pipes' just go into the one flange and onto the exhaust.

the 4-1 manifold is better for high rev range use as it looses a bit of torque at mid range
4-2-1 is the better of the 2 tbh more torque mid range and the same at high rpm's
