I have my Iphone unlocked using Ziphone, apparantly Iphone 2.0 is available now. Can I just update it using Itunes or will that just lock it again?

Don't update it!
Mate did this whilst it was unlocked and it's now blocked.
I am just not updating it, but if you want too, you'd have to wipe the phone, update it then re-jail break it. Far too much hassle.

I've read the 2.0 break is only available for MAC users at the mo

Not much I don't think, you get the Appstore and a couple of other things I think. I was just gona do it if it was an easy job, which its seeming like it won't be so I probably won't bother.
