Right, I have just bought a 32GB pendrive and I am trying to move a 4gb movie over to it from my self powered portable hard drive......it comes up with "file is too large for destination folder"
The USB pen is brand new and is empty! Is there a setting that needs to be changed so I can copy larger files over?!

Did you get the 32gb pen drive from ebay / china? I've bought them before and in reality theyre like 512mb.
If not, as said, format it.

Think so, format to NTFS and it should work.

If it's for your TV then note that most don't support the NTFS file system only FAT based.
